"The former owner was so pleased that the name would continue and encouraged us to set up our business as Mt. Pilchuck Ski & Sport," said Lynn Marks, CEO of the business she owns with her husband, Bentley. The couple lives in Arlington.

Opened a few weeks ago on 172nd Street NE near the Arlington Airport, the store sells its own line of skis and snowboards, plus jackets, hats and other winter sports products and accessories. There's a table-and-chair lounging area styled as a mountain cabin, plus hand-made products from area residents, including knitted hats and beaded bracelets.

Behind the launch of the Marks' Mt. Pilchuck Ski & Sport venture was the Markses' dream of promoting their patented invention, the iCycle Yeti, that features a bicycle-like frame with handlebars and snowboards instead of wheels.

It's been a long struggle for the Markses to see their downhill snow scooter become popular. The Yeti blends the attractions of snowboarding and biking to create a new snow experience.

Eleven years ago in Snohomish, Bentley Marks watched his niece ride a Razor scooter and began thinking about creating something similar for the snow sport industry.

"The first one built in my garage weighed 69 pounds," said Bentley Marks, a former airline pilot. "When we tested it in Nevada, we proved the concept worked but it shook when it went too fast. One fellow who looked at it helped us re-engineer the frame and an engineer doing freelance CAD work helped to fine-tune it."

After producing a much lighter version and finding a manufacturer, the couple founded The Slider Corp. to market the Yeti and began promoting it, attracting the attention of the national television program "Heartbeat of America," hosted by William Shatner. Their website also notes publicity on KING-TV's "Evening Magazine," in Ski Area Management magazine, online at Skipressworld.com and in local publications.

"My plan hasn't changed," he said. "I'm on track with what the doctors have told me to do, and I'm doing that. I'm rehabbing hard."

The NFL would prefer to keep the focus on this week's juiciest story lines -- Giants quarterback Eli Manning trying to outdo his older brother by winning a second Super Bowl; Patriots quarterback Tom Brady and coach Bill Belichick chasing their fourth titles and maybe exacting a little revenge for their loss to New York four years ago.

But as everyone knows, this is Peyton's place, his town, and fans hang on every detail of his recovery.

"It's not the way it should be," he said earlier Tuesday in a taped interview with ESPN.

The Colts ended their most tumultuous month in more than a decade with another flurry of moves.

Team officials confirmed Tuesday that they had hired Bruce Arians, Manning's former quarterbacks coach, as offensive coordinator, and Harold Goodwin as offensive line coach. They also fired tight ends coach Ricky Thomas and assistant offensive line coach Ron Prince.

The whirlwind of changes began Jan. 2, the day after the Colts clinched the No. 1 overall draft pick with a league-worst 2-14 record. Irsay has since fired vice chairman Bill Polian, general manager Chris Polian, and dismissed coach Jim Caldwell and most of his staff -- a series of changes that prompted Peyton Manning to voice his complaints two days after the conference championship games.

Two days later, Irsay called Manning a "politician," fueling a public spat that drew national attention. The two tried to put out that fire before the team's scheduled arrivals, but the fallout has continued into this week.

"I would think he's very uncomfortable, especially with the Giants here and Eli," former Colts coach Tony Dungy said. "When you get to a Super Bowl, even though it's the second (recent) one for the Giants, you want the focus to be on that achievement. He would much rather have the focus on Eli and what they've done to get here, and unfortunately, that's not going to happen."

If there were any doubts, they were answered in a strange 24-hour period that began with the host committee's news conference Monday.

Irsay joined the mayor, governor and two committee officials to discuss game-week preparations, but wound up taking most of the questions, nearly all of which focused on Manning's future. Afterward, Irsay said he didn't plan to discuss the Manning situation again this week.

On Tuesday, Manning piped in.

Shortly after the NFL's annual Super Bowl media day event ended, Peyton Manning met with a small group of reporters with the idea of turning the attention back to Sunday's game.

That didn't work too well, either.

"I'm working hard, I had a really good session today," Manning said after repeatedly being asked about his rehab. "I continue to make progress and work hard. The doctors are encouraged and that's encouraging to me."

The four-time league MVP hasn't played in more than a year because of a damaged nerve that caused weakness in his throwing arm. He had neck surgery in May, then underwent his third and most invasive neck surgery in 19 months in September. Doctors fused two vertebrae together, a procedure that forced him to miss the Colts two-win season.

Since then, there has been rampant speculation about his recovery, the potential risks of a return, whether the Colts will pay Manning a $28 million roster bonus in early March to prevent him from becoming a free agent or whether the soon-to-be 36-year-old might retire.

"My plan hasn't changed," Manning said. "I'm on track with what the doctors have told me to do, and I'm doing that. I'm rehabbing hard."

About one in five or six cases of adult leukaemia in Western populations relates to so-called chronic myeloid leukaemia, or CML. Treatment of CML usually relies on inhibitors of the abnormal protein that causes the condition but some patients do not respond to treatment and efforts are underway to develop a supplementary approach, targeting the so-called JAK2 kinase. Recent results from the groups of Veronika Sexl at the University of Veterinary Medicine, Vienna (Vetmeduni Vienna) and Giulio Superti-Furga at the Research Center for Molecular Medicine of the Austrian Academy of Sciences (CeMM) have called this strategy into question. The work is published in the current issue of the prestigious journal Nature Chemical Biology and is of immediate relevance to leukaemia treatment.

The cause of CML has been known since 1960, when two scientists in Philadelphia, Pennsylvania showed that the disease was associated with a particular genetic abnormality, the "Philadelphia chromosome". Philadelphia chromosomes represent the result of an incorrect crossing-over between two chromosomes, through which part of the Bcr ("breakpoint cluster region") gene from chromosome 22 fuses with the Abl gene on chromosome 9. The fusion gene product is a tyrosine kinase, i.e. it can phosphorylate other proteins on tyrosine residues. When it does so, it incorrectly activates several signal pathways controlling cell division in white blood cells and leads to leukaemia. Thankfully, drugs have been developed that prevent the kinase activity of the BCR-ABL fusion protein and the majority of patients treated with such tyrosine kinase inhibitors (e.g. imatinib) show no further signs of leukaemia.

Unfortunately, however, patients may develop resistance to the therapy so an alternative approach is required. Recent developments have focused on the use of drugs targeting another kinase involved in CML, the JAK2 kinase. In normal white blood cells, JAK2 is known to activate a further molecule, known as STAT5, which is absolutely required for the development of CML. The argument runs that if JAK2 could be specifically inhibited ? and thus STAT5 not activated ? it would bring fresh hope to patients who do not respond to treatment with imatinib. Several potential inhibitors of JAK2 are currently undergoing clinical trials and may shortly be available for treating patients.

The theory is appealing but to date we do not really understand exactly what happens when JAK2 is inactivated following the initiation of leukaemia by the Bcr-Abl oncogene. Sexl and her colleagues have used a transgenic mouse model to clarify the functions of these proteins in leukaemia. Their results were highly unexpected. The JAK2 kinase was found to be not required for the maintenance of the disease, i.e. inhibiting JAK2 in leukaemic cells had no therapeutic benefit. However, inhibition of STAT5 in leukaemia was sufficient to prevent cell proliferation. As Sexl says, "this means that the normal signalling pathway is completely rewired in CML cells: STAT5 activity no longer depends on JAK2." In support of this conclusion, the researchers were able to show that the BCR-ABL protein directly phosphorylates STAT5, thereby activating it.

As Superti-Furga notes, "The findings have extremely important consequences for CML therapy in humans," adding, "We are very happy that the collaboration between our two groups is so fruitful". Put bluntly, leukaemia patients that do not respond to imatinib will not be helped by inhibiting JAK2. Interestingly, some JAK2 inhibitors do slow the progression of leukaemic cells, although they must be given at very high levels. The "therapeutic" action is mediated by a secondary target of the JAK2 inhibitors, which Sexl and colleagues have shown to be the Bcr-Abl oncogene itself. Sexl concludes that, "at the moment there is simply no rationale for giving leukaemic patients JAK2 inhibitors. If we want to help patients who do not respond to imatinib, we should concentrate instead on developing inhibitors to STAT5."

Few in baseball were surprised that two well-established players had misrepresented themselves. The fear is that the problem could be much more widespread. One agent said more than a dozen players could soon lose their contracts because of age and identity issues.

?These are like time bombs,? Mark Newman, the Yankees? senior vice president for baseball operations, said by telephone from the Dominican Republic while scouting there last week.

For his part Newman, as well as others in baseball, believe that the problem will get better. Still: as long as there are millions to be gained by an 18 or 19 year-old passing himself off as 16, this problem is going to persist.
